=== Theoretical === The theory of general chromatography applies to HPLC. This theory is the basis for system-suitability tests of the United States Pharmacopoeia, which are a set of quantitative criteria for how suitable a HPLC system is to the required analysis at any step of it. This relation is also represented as a normalized unit-less factor known as the retention factor, or retention parameter, which is the experimental measurement of the capacity ratio, as shown in the Figure of Performance Criteria as well. tR is the retention time of the specific component and t0 is the time it takes for a non-retained substance to elute through the system without any retention, thus it is called the Void Time. Polystyrene ion exchangers with large pores can be used for the separation of protein but must be coated with a hydrophilic substance. Cellulose based medium can be used for the separation of large molecules because they contain large pores. Protein binding in this medium is high and has low hydrophobic character. DEAE is an anion exchange matrix that is produced from a positive side group of diethylaminoethyl bound to cellulose or Sephadex. Agarose gel based medium contain large pores as well but their substitution ability is lower in comparison to dextrans.

=== Negative effects === As of 2017, the American Academy of Pediatrics says that fruit juice should not be given to children under the age of one due to its lack of nutritional benefit. For children ages one to six, intake of fruit juice should be limited to less than 4–6 oz (110–170 g) per day (about a half to three-quarters of a cup) due to its high sugar and low fiber content compared to fruit. Overconsumption of fruit juices may reduce nutrient intake compared to eating whole fruits and may produce diarrhea, gas, abdominal pain, bloating, or tooth decay. Overconsumption of fruits and fruit juice may contribute to dental decay and cavities via the effect of fruit acids on tooth enamel. Longitudinal prospective cohort studies showed a significantly increased risk of type 2 diabetes when juices with added sugars were consumed compared to eating whole fruits. A 2014 review found that a higher intake of sugar-sweetened fruit juice was significantly associated with a higher risk of type 2 diabetes. Overconsumption of fruit juice with added sugars has also been linked to childhood obesity. The American Journal of Public Health proposed that the Healthy Hunger-Free Kids Act of 2010 in the United States eliminate 100% fruit juices and substitute them instead with whole fruits.

=== Diagnosis === The assessment of vitamin B6 status is essential, as the clinical signs and symptoms in less severe cases are not specific. The three biochemical tests most widely used are plasma PLP concentrations, the activation coefficient for the erythrocyte enzyme aspartate aminotransferase, and the urinary excretion of vitamin B6 degradation products, specifically urinary PA. Of these, plasma PLP is probably the best single measure, because it reflects tissue stores. Plasma PLP of less than 10 nmol/L is indicative of vitamin B6 deficiency. A PLP concentration greater than 20 nmol/L has been chosen as a level of adequacy for establishing Estimated Average Requirements and Recommended Daily Allowances in the USA. Urinary PA is also an indicator of vitamin B6 deficiency; levels of less than 3.0 mmol/day is suggestive of vitamin B6 deficiency.
