RP-HPLC raises a handful of sensible questions. This page answers them in order, starting with the fundamentals and moving to applications.
This page was last updated on 2026-05-15 and is reviewed periodically as new material appears.
The peptide is prone to several degradation pathways. Oxidation of the methionine residue produces a sulfoxide that elutes close to the parent peak in many chromatographic systems. Hydrolysis of peptide bonds and deamidation of susceptible residues in related sequences also reduce purity over time. Lyophilised material kept dry at minus twenty degrees Celsius and shielded from light is the most stable form commonly described in laboratory practice.
Material sold for laboratory use varies widely in stated purity and documentation. A certificate of analysis should list the analytical method, the column and detector used, and the observed purity value. Independent verification by an outside laboratory is the practical way to confirm identity when documentation is absent or internally inconsistent. Regulatory status differs by country, and a product legal in one jurisdiction may be unapproved or controlled in another.

Stability depends heavily on physical state. Lyophilized powder held dry, cold and dark retains its content over long periods, whereas dissolved peptide begins to change within days at room temperature. The most cited degradation route is oxidation of the methionine residue, which converts the peptide to a sulfoxide form that elutes differently on chromatography. Hydrolysis of amide bonds and adsorption onto container walls contribute smaller losses. Buffers that exclude oxygen from the headspace slow the oxidation pathway, but no single condition prevents all change indefinitely.
Practical handling follows from those properties. Bulk material is best divided into single-use portions soon after receipt, because each thaw exposes the whole container to moisture and temperature cycling. Vials should be allowed to reach room temperature before opening to prevent condensation on the powder. Low-binding plasticware reduces loss of dilute solutions, and sterile filtration is used when a preparation must remain free of microbial growth. Records of batch number, reconstitution date and storage history are what allow a later analytical result to be interpreted meaningfully.
Identity and purity of Semax are established mainly by reversed-phase high-performance liquid chromatography coupled with mass spectrometry. The chromatographic trace gives a purity estimate as a percentage of total peak area, while electrospray or matrix-assisted laser desorption ionization confirms the molecular mass against the calculated value. Amino acid analysis and sequence-specific fragmentation provide further confirmation when a supplier's chain of custody is unclear. Vendors frequently quote a purity figure without stating the detection wavelength or the integration method, which limits how far one number can be compared with another.
Later generations of the molecule include an N-acetylated form and an amidated form, both marketed online alongside the parent peptide. These variants differ in terminal chemistry and stability, and they are frequently discussed in the same breath even though they have not been compared in controlled trials. Supply outside formal healthcare systems comes largely from laboratories that synthesize peptides to order. Purity and identity of these materials vary widely, and no single body oversees the international trade.
Semax is a synthetic heptapeptide developed in the Soviet Union during the 1980s by researchers at the Institute of Molecular Genetics in Moscow. It was designed as a truncated analog of adrenocorticotropic hormone, retaining only the fragment spanning residues four through ten. Investigators sought a peptide that would preserve the cognitive effects associated with ACTH while eliminating the hormonal stimulation of the adrenal cortex. The compound entered clinical use in Russia during the following decade.
Russian regulatory authorities approved the peptide for nasal administration, and it remains listed in the national pharmacopoeia under several trade names. Documented indications include acute ischemic stroke, transient ischemic attacks, traumatic brain injury, and certain ophthalmological and neurological conditions. Physicians also prescribe it for cognitive complaints in older patients, although the evidence base for that use is thinner. Outside Russia and a few neighboring states, the substance is not an approved medicine and is sold instead as a research chemical.

The first death row prisoner to be hanged in Singapore during the COVID-19 pandemic was 68-year-old Singaporean drug offender Abdul Kahar Othman on 30 March 2022, who had not appealed against his sentence and later executed as scheduled, therefore resuming executions in Singapore. By the time Abdul Kahar was executed, there were 62 prisoners on death row, awaiting execution (reduced to 61 with Nagaenthran's execution). Nagaenthran K. Dharmalingam was the second to be hanged on 27 April 2022 after Abdul Kahar. Datchinamurthy Kataiah was originally the third in line to be executed on 29 April 2022 after Nagaenthran before his execution was postponed due to an appeal. On 7 July 2022, Kalwant Singh Jogindar Singh and Norasharee Gous became the third and fourth convicts to be hanged in Singapore in 2022. In the same month, Nazeri Lajim was executed 15 days after Kalwant and Norasharee. Three more hangings - one on 26 July and two (Malaysian Rahmat Karimon and his accomplice Zainal Hamad) on 2 August - were conducted after Nazeri's execution. On 5 August 2022, 45-year-old Singaporean Abdul Rahim Shapiee and his 49-year-old accomplice Ong Seow Ping were the ninth and tenth to be executed. A 55-year-old Singaporean, whose name is unknown, was the eleventh to be executed for a drug charge on 7 October 2022. In total, eleven executions, all for drug trafficking, took place in the year 2022. In 2023, the first execution was carried out on 26 April 2023, when a 46-year-old Singaporean Tangaraju Suppiah, who was convicted in 2018 of marijuana trafficking, was hanged at dawn.
== Mechanotaxis in development == Cell migration is essential in early embryonic development, as a defining characteristic of this phase is the folding and reorganization of the embryo that occurs during and after gastrulation. Without cell migration, complex structures involving multiple cell types that make up complex organisms – like tissues, organs, limbs, etc. – would not develop correctly. There are multiple factors that influence cells to move during development –– but the factors that influence mechanotaxis in development often involve interactions between cells or between a cell and a substrate such as a yolk or membrane. Contact inhibition of locomotion is involved in the migration of many cell types, including neural crest (NC) cells in vertebrates which give rise to cells of the peripheral nervous system (PNS), facial cartilage, and other non-neural cells throughout the body. NC cells are very mobile, with actin-rich protrusions at the leading edge of each cell in the direction of travel. When an NC cell collides with another NC cell, activation of the Wnt planar cell polarity (PCP) signaling pathway occurs at the point of cell contact, causing localized activation of the downstream effector RhoA. This activation is likely caused by interactions between cadherins on the cell surfaces, and leads to the retraction of the cell protrusions and a change in the cell's polarity, causing the NC cell to change direction.

Inflammation (from Latin: inflammatio) is part of the biological defence response of body tissues. Inflammatory immunovascular responses can be triggered by a broad range of stimuli, including physical trauma, "dead, damaged, malfunctioning or stressed tissues", pathogens, irritants, toxins, overuse, autoimmunity, allergens, and foreign bodies (e.g. silica and asbestos). The five cardinal signs are heat, pain, redness, swelling, and loss of function (Latin calor, dolor, rubor, tumor, and functio laesa). Inflammation is a generic response, and therefore is considered a mechanism of innate immunity, not adaptive immunity. It involves immune cells, blood vessels, and molecular mediators. The function of inflammation is to eliminate the initial cause of cell injury, clear out damaged cells and tissues, and initiate tissue repair. Too little inflammation can lead to progressive tissue destruction by the harmful stimulus (e.g. bacteria) and compromise the survival of the organism, but too much inflammation, in the form of chronic inflammation, is associated with various diseases, such as hay fever, periodontal disease, atherosclerosis, and osteoarthritis. Inflammation can be classified as acute or chronic. Acute inflammation is the initial response of the body to harmful stimuli, and is achieved by the increased movement of plasma and leukocytes (in particular granulocytes) from the blood into the injured tissues.
=== Human === In humans, ADH exists in multiple forms as a dimer and is encoded by at least seven genes. Among the five classes (I-V) of alcohol dehydrogenase, the hepatic forms that are used primarily in humans are class 1. Class 1 consists of α, β, and γ subunits that are encoded by the genes ADH1A, ADH1B, and ADH1C. The enzyme is present at high levels in the liver and the lining of the stomach. It catalyzes the oxidation of ethanol to acetaldehyde (ethanal):
The independence of Poland had been campaigned for in Russia and in the West by Dmowski and in the West by Ignacy Jan Paderewski. Tsar Nicholas II of Russia, and then the leaders of the February Revolution and the October Revolution of 1917, installed governments who declared in turn their support for Polish independence.[d1] In 1917, France formed the Blue Army (placed under Józef Haller) that comprised about 70,000 Poles by the end of the war, including men captured from German and Austrian units and 20,000 volunteers from the United States. There was also a 30,000-men strong Polish anti-German army in Russia. Dmowski, operating from Paris as head of the Polish National Committee (KNP), became the spokesman for Polish nationalism in the Allied camp. On the initiative of Woodrow Wilson's Fourteen Points, Polish independence was officially endorsed by the Allies in June 1918.[c1] In all, about two million Poles served in the war, counting both sides, and about 400–450,000 died. Much of the fighting on the Eastern Front took place in Poland, and civilian casualties and devastation were high.
In 2009, Chipotle released an app for the iPhone that allows users to find nearby Chipotle locations, place an order, and prepay with a credit card. In 2013, Chipotle released an Android app that allows users to locate nearby Chipotle locations, place an order, prepay with a credit or gift card, and access favorites and recent orders. In late 2021, the company added a non-food item to the menu—cilantro soap. According to published reports, the soap sold out the day after its release. The soap was produced after an August 2021 Instagram post of what was thought to be a mock photo of the product. It was an effort to engage fans and haters of the herb that, "plays into a larger trend of turning digital moments into real life experiences," according to Chris Brandt, Chipotle's Chief Marketing Officer. Chipotle uses green marketing and its "Food with Integrity" platform to boost its brand image.

In 2014, Black Mesa was named by PC Gamer among the "Ten top fan remade classics you can play for free right now". The remake's final release in 2020 was similarly praised by reviewers. On OpenCritic, Black Mesa was recommended by 100% of critics, based on 15 reviews. Writing for the UK PC Gamer, Andy Kelly said the project felt like a professional work. He found that while the original Half-Life structure hampered some of Black Mesa, the redesigns worked well, particularly the new Xen sections, which he felt provided more closure than the original. Eurogamer said that Black Mesa felt more like an evolution than a remake of Half-Life, with the designs of the Crowbar Collective to trim down certain levels while adding other features helped to enhance the overall product, making the overall title more of a survival horror than a first-person shooter. Dario Casali, a designer at Valve who has worked on all Half-Life games, remarked in an interview that during development on Half-Life: Alyx he attempted to play the entirety of the original Half-Life again for research, but after five hours decided to play Black Mesa instead, reasoning it was a much more enjoyable product.
Emicerfont (GW-876,008) is a drug developed by GlaxoSmithKline which acts as a CRF-1 antagonist. Corticotropin releasing factor (CRF), also known as Corticotropin releasing hormone, is an endogenous peptide hormone which is released in response to various triggers such as chronic stress, and activates the two corticotropin-releasing hormone receptors: CRF1 and CRF2. This then triggers the release of corticotropin (ACTH), another hormone which is involved in the physiological response to stress. Emicerfont blocks the CRF1 receptor, and so reduces ACTH release. It has been investigated for the treatment of irritable bowel syndrome (IBS) and alcoholism, and while it was not effective enough to be adopted for medical use in these applications, it continues to be used for research, as the role of the CRH-ACTH system in IBS remains poorly understood.
For the following year, Ubayd Allah was bogged down in battles with the Qaysi tribes of Jazira led by Zufar ibn al-Harith al-Kilabi. By 686, Ubayd Allah's army numbered some 60,000 troops. By the time Ubayd Allah's army approached Mosul toward Iraq, the Zubayrids under Mus'ab ibn al-Zubayr had established themselves in Basra while al-Mukhtar ibn Abi Ubayd took control of Kufa in the name of the Alid Muhammad ibn al-Hanafiyya. Al-Mukhtar dispatched Ibrahim ibn al-Ashtar and an army composed largely of non-Arab freedmen to confront Ubayd Allah. The latter fended off the first wave of al-Mukhtar's troops, and proceeded to face off Ibn al-Ashtar at the Khazir River. In the ensuing Battle of Khazir, the Umayyad army was routed and Ubayd Allah was slain by Ibn al-Ashtar. His lieutenants Husayn, Shurahbil and al-Rabi'a were also killed. With Ubayd Allah's death, Caliph Abd al-Malik halted further advances against Iraq until 691.
=== Familial dysautonomia === Familial dysautonomia is a genetic condition that can be associated with a lack of overflow tears (alacrima) during emotional crying. Obstruction of the punctum, nasolacrimal canal, or nasolacrimal duct can cause even normal levels of the basal tear to overflow onto the face (epiphora), giving the appearance of constant psychic tearing. This can have significant social consequences.
He proposed that a World Peace Research Organization be set up as part of the United Nations to "attack the problem of preserving the peace". Pauling also supported the work of the St. Louis Citizen's Committee for Nuclear Information (CNI). This group, headed by Barry Commoner, Eric Reiss, M. W. Friedlander and John Fowler, organized a longitudinal study to measure radioactive strontium-90 in the baby teeth of children across North America. The "Baby Tooth Survey", published by Louise Reiss, demonstrated conclusively in 1961 that above-ground nuclear testing posed significant public health risks in the form of radioactive fallout spread primarily via milk from cows that had ingested contaminated grass. The Committee for Nuclear Information is frequently credited for its significant contribution to supporting the test ban, as is the ground-breaking research conducted by Reiss and the "Baby Tooth Survey". Public pressure and the frightening results of the CNI research led to a moratorium on above-ground nuclear weapons testing, followed by the Partial Test Ban Treaty, signed in 1963 by John F. Kennedy and Nikita Khrushchev. On the day that the treaty went into force, October 10, 1963, the Nobel Prize Committee awarded Pauling the Nobel Peace Prize for 1962.

Reversed-phase high-performance liquid chromatography is the standard method, with detection in the ultraviolet range. Peak area percentage yields a purity figure for the main component. Mass spectrometry is normally run alongside to confirm molecular identity.
Dry lyophilised powder is generally kept at minus twenty degrees Celsius, protected from light and moisture. Repeated freeze-thaw cycles are avoided because they encourage aggregation and moisture uptake. Solutions are less stable than the solid form and degrade faster at room temperature.
A certificate documents what the supplier measured, not what an independent party confirmed. Methods, instruments and acceptance criteria can differ between suppliers. Third-party testing is the usual way to resolve discrepancies.
