Once dissolved, the peptide is considerably less stable than the solid. Aqueous solutions are usually prepared at neutral to slightly acidic pH, filtered, and divided into single-use aliquots before freezing. Repeated freeze-thaw cycles are a common cause of avoidable loss and are best prevented by never refreezing a thawed aliquot. Adsorption to plastic and glass surfaces can lower the measured concentration of dilute solutions, particularly below roughly 0.1 mg/mL. Buffer choice, salt content, and container material all influence how much peptide remains detectable after storage.
Identity and purity are established with standard peptide methods. Reversed-phase HPLC with ultraviolet detection near 214 nm resolves the parent peak from deletion and truncation byproducts, and reports typically quote a main-peak percentage. Mass spectrometry by electrospray or MALDI-TOF confirms the expected molecular mass, while amino acid analysis or peptide mapping can verify composition when the sequence itself is in question. A certificate of analysis that pairs a chromatogram with a mass spectrum is more informative than a purity figure alone. Counter-ion content and residual solvents are separate specifications and are frequently omitted.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its design combines the ACTH(4-7) core fragment with a C-terminal Pro-Gly-Pro extension, a modification intended to improve stability and prolong activity. The molecule is hydrophilic, carries no lipid chains or glycosylation, and has a theoretical mass just over 810 daltons in its free form. All seven residues are proteinogenic amino acids, so no non-natural building blocks appear in the backbone. A free N-terminal methionine and C-terminal proline define the unmodified parent peptide.
The compound was developed during the 1980s at the Institute of Molecular Genetics in Moscow as part of research on fragments of adrenocorticotropic hormone. Early work examined short ACTH-derived sequences that retained neurotrophic effects while lacking the endocrine activity of the full hormone. Semax entered clinical use in Russia during the 1990s, where it received registration for several neurological indications. Outside that region it remained primarily a laboratory research material rather than an approved therapeutic. English-language literature on it grew more slowly and frequently cited the original Russian studies.

Identity and purity are confirmed with reversed-phase high-performance liquid chromatography, typically monitored at 214 nanometres where the peptide bond absorbs. Mass spectrometry, either electrospray or MALDI-TOF, verifies molecular mass against the theoretical value and detects truncation or adduct formation. Amino acid analysis and peptide mapping provide additional confirmation when required. The most frequently reported impurities are deletion sequences from incomplete coupling, methionine sulfoxide from oxidation, and dimeric species formed through non-covalent aggregation. Impurity profiles depend strongly on the synthesis and purification route chosen by the producer.
Published research has focused mainly on neurological and cognitive endpoints in animal models, with proposed mechanisms involving brain-derived neurotrophic factor and related signalling pathways. A substantial share of the human data originates from a limited number of research groups, and independent replication in other countries remains sparse. Regulatory status reflects that distribution: the peptide is registered as a medicine in Russia and appears in some neighbouring markets, while elsewhere it is handled as a research chemical without approved therapeutic labelling. Questions about dose-response relationships, long-term effects, and comparability across studies are still open.
Lyophilised powder is normally kept at -20 °C in a desiccated container, with some suppliers recommending -80 °C for long-term archival storage. Repeated freeze-thaw cycles are the most common cause of avoidable loss, so aliquoting before freezing reduces variability between working sessions. Dissolved peptide is far less stable than the dry solid and is usually prepared fresh or held briefly at 4 °C. Aqueous solutions support both hydrolysis of the backbone and oxidation of the N-terminal methionine, and these two routes dominate degradation under ordinary laboratory conditions.
Pharmacological accounts link semax to melanocortin signalling and to modulation of neurotrophic factor expression, particularly brain-derived neurotrophic factor and nerve growth factor. Much of this evidence comes from rodent studies using intranasal delivery, a route chosen because it allows peptides to reach the central nervous system with limited systemic exposure. Whether the same mechanisms operate in humans at comparable magnitude remains an open question. The precise receptor or receptors responsible for the reported behavioural and neuroprotective effects have not been conclusively identified.
Clinical reports describe use in ischaemic stroke, transient ischaemic attack, optic nerve conditions, and cognitive complaints, but most of these studies are small and were conducted in a single region. Systematic reviews have generally described the evidence base as limited in size and variable in methodological quality. Randomised controlled data suitable for international regulatory assessment are scarce. As a result, major treatment guidelines outside Russia do not include the peptide, and interest in it remains largely research-driven rather than routine clinical.
Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. It is described in the literature as an analogue of the ACTH(4–10) fragment, a short N-terminal portion of adrenocorticotropic hormone that retains some neurotropic activity without the full hormonal effects of the parent peptide. The molecule carries a methionine residue at the N-terminus and two proline residues near the C-terminus, features that shape both its interactions with receptor systems and its chemical stability. The free peptide corresponds to the formula C37H51N9O10S and a molecular mass near 813.9 Da.
Dissolution is normally performed in water, phosphate-buffered saline, or normal saline, since the peptide is freely soluble in aqueous media and is rarely handled with strong organic solvents. Solution pH should be kept near neutral, because extreme acidity or alkalinity accelerates backbone hydrolysis and encourages oxidation of the methionine side chain. Once dissolved, the material is less stable than the dry powder and is commonly divided into single-use portions and frozen. Buffers containing primary amines, such as Tris, are often avoided because of possible side reactions.
Purity assessment relies mainly on reversed-phase high-performance liquid chromatography, which separates the target heptapeptide from truncated sequences, deletion analogues, and oxidised forms. Mass spectrometry, usually coupled to liquid chromatography, confirms identity through the expected molecular ion and reveals modifications such as methionine sulfoxide formation. Amino acid analysis can verify composition, and tandem mass spectrometry supports sequence confirmation. Ultraviolet detection near 254 to 280 nanometres is convenient because the phenylalanine and histidine residues absorb in that region. Nuclear magnetic resonance is rarely used for routine release testing.
This may be due to the presence of a specific estramustine-binding protein (EMBP) (Kd = 10–35 nM for estramustine), also known as prostatin or prostatic secretion protein (PSP), which has been detected in prostate cancer, glioma, melanoma, and breast cancer cells. Because of its tissue selectivity, EMP is said to produce minimal cytostatic effects in healthy tissues, and its tissue selectivity may be responsible for its therapeutic cytostatic efficacy against prostate cancer cells. EMP was originally developed as a dual ester prodrug of an estrogen and normustine as a nitrogen mustard alkylating antineoplastic agent which, due to the affinity of the estrogen moiety for estrogen receptors, would be selectively accumulated in estrogen target tissues and hence estrogen receptor-positive tumor cells. Consequentially, it was thought that EMP would preferentially deliver the alkylating normustine moiety to these tissues, allowing for reduced cytostatic effects in healthy tissues and hence improved efficacy and tolerability. However, subsequent research found that there is very limited and slow cleavage of the normustine ester and that EMP is devoid of alkylating activity. In addition, it appears that estramustine and estromustine may be preferentially accumulated in estrogen target tissues not due to affinity for the estrogen receptors, but instead due to affinity for the distinct EMBP. Extremely high, pregnancy-like levels of estradiol may be responsible for the leukocytosis (increased white blood cell count) that is observed in individuals treated with EMP.

=== Ethylene === Pyrolysis is used to produce ethylene, the chemical compound produced on the largest scale industrially (>110 million tons/year in 2005). In this process, hydrocarbons from petroleum are heated to around 600 °C (1,112 °F) in the presence of steam; this is called steam cracking. The resulting ethylene is used to make antifreeze (ethylene glycol), PVC (via vinyl chloride), and many other polymers, such as polyethylene and polystyrene.

===== Funding freezes ===== From the outset of 2025, NIH funding operations have faced interruptions on an unprecedented scale under the direction of the current executive branch of the U.S. government; disruptions as of March 2025 include the following: • impeding grants for dementia and ALS research; • hindering procurement of necessary resources, such as those for transporting patient blood samples; • preventing a research scientist from consulting with physicians treating children with a devastating rare condition; • interrupting the supply of mice for genetic studies, with years of research being imperiled as a result; • cutting research grants for training doctoral and postdoctoral students. This has led to protests such as the Bethesda Declaration, an open letter from former and current NIH staffers.

Rivastigmine, sold under the brand name Exelon among others, is an acetylcholinesterase inhibitor used for the treatment of dementia associated with Alzheimer's disease and with Parkinson's disease. Rivastigmine can be administered orally or via a transdermal patch; the latter form reduces the prevalence of side effects, which typically include nausea and vomiting. Rivastigmine is eliminated through the urine, and appears to have relatively few drug-drug interactions. It was patented in 1985 and came into medical use in 1997.
=== Trichotillomania === High doses of inositol have been explored for treatment of trichotillomania (compulsive hair-pulling) and related disorders, but no definitive evidence points to its effectiveness.
In the event of involvement of the sympathetic tracts in the brainstem, a partial Horner's syndrome may develop; this is the combination of a drooping eyelid, constricted pupil, and an apparently sunken eye on one side of the face. If the dissection of the artery extends to the part of the artery that lies inside the skull, subarachnoid hemorrhage may occur (1% of cases). This arises due to rupture of the artery and accumulation of blood in the subarachnoid space. It may be characterized by a different, usually severe headache; it may also cause a range of additional neurological symptoms. 13–16% of all people with vertebral or carotid dissection have dissection in another cervical artery. It is therefore possible for the symptoms to occur on both sides, or for symptoms of carotid artery dissection to occur at the same time as those of vertebral artery dissection. Some give a figure of multiple vessel dissection as high as 30%.
Both malate and oxaloacetate can be converted into phosphoenolpyruvate, which is the product of phosphoenolpyruvate carboxykinase, the first enzyme in gluconeogenesis. The net result of the glyoxylate cycle is therefore the production of glucose from fatty acids. Succinate generated in the first step can enter into the citric acid cycle to eventually form oxaloacetate.
In an open letter to the United Kingdom government the British Society for Middle Eastern Studies (BRISMES) called the perceived systematic targeting of universities, schools, laboratories and libraries as a component of a genocidal strategy aimed at destroying the Palestinian educations system in Gaza. The chair of the Committee on the Rights of the Child stated on 8 February, "The rights of children living under the state of Israel’s effective control are being gravely violated at a level that has rarely been seen in recent history." The Gulf Cooperation Council called on the UN Human Rights Council to "shoulder their responsibility in ending the tragedy of the children in Gaza and securing their right to live, safety, and protection". The UN Population Fund stated in late-March 2024 that "Pregnant women and new mothers face a constant struggle to keep themselves and their newborns alive." In response to reports that one-third of under-two-year-olds in northern Gaza were acutely malnourished, UNICEF's James Elder, stated, "That is a terrifying number". The UN added Israel to a blacklist of countries that violate the rights of children. In June 2024, the United Nations stated that grave violations against children in conflict zones had increased 155 percent globally, due especially to explosive weapons used in the Gaza Strip. In September 2024, a UN committee stated Israel had committed severe breaches against children's rights in Gaza, saying, "The outrageous death of children is almost historically unique. This is an extremely dark place in history.

Each freeze-thaw cycle can degrade a fraction of the peptide and promote aggregation. Dividing a stock solution into single-use volumes removes repeated cycling as a variable. It also limits the time a thawed solution spends at room temperature.
A high main-peak percentage says little about identity, counter-ion content, or residual solvents. It also does not distinguish an isomer or a closely eluting analog. Mass confirmation and the accompanying documentation cover those gaps.
Salt form changes mass, solubility, and hygroscopicity, and acetate and trifluoroacetate salts behave differently in solution. Storage guidance still centers on -20 °C for powder and lower temperatures for solutions. The certificate should state which salt is present.
It is a synthetic seven-amino-acid peptide built from the ACTH(4-7) sequence plus a Pro-Gly-Pro tail. It belongs to the family of short ACTH fragments studied for nervous-system effects. Its backbone contains only standard amino acids.
