Reported pharmacological work centers on neurotrophic signaling, including changes in BDNF and NGF expression in hippocampal tissue in animal models. Human data come largely from studies conducted in Russia, and how well those results generalize to other populations remains an open question. Regulatory status differs sharply by jurisdiction: Semax is a registered prescription medicine in Russia, while it holds no approved marketing status in the United States or the European Union. Outside such jurisdictions it is generally handled as a research chemical, which affects both documentation and quality expectations.
Semax is a synthetic heptapeptide whose sequence is Met-Glu-His-Phe-Pro-Gly-Pro. It was developed as a fragment analog of adrenocorticotropic hormone, modeled specifically on the ACTH(4-10) region. The first four residues reproduce that fragment, while a Pro-Gly-Pro tripeptide is appended at the C-terminus. Work on the compound originated in Russia, where it entered clinical use as an intranasal preparation. Its sequence places it among short regulatory peptides studied for effects on the central nervous system rather than on the adrenal axis.
The C-terminal Pro-Gly-Pro extension is not incidental. Proline-rich tails are known to resist several common peptidases, and the published literature attributes the longer half-life of Semax, relative to unmodified ACTH fragments, to this feature. The modification also removes the melanocyte-stimulating and corticosteroidogenic activity that characterizes longer ACTH-derived sequences. Because the molecule is small and hydrophilic, it is typically formulated as an aqueous solution for intranasal or parenteral delivery. Acetylation or amidation at the termini appears in closely related research peptides and shifts the mass by a fixed increment.

Semax is a synthetic heptapeptide with the sequence Met-Glu-His-Phe-Pro-Gly-Pro. Its chain combines the first seven residues of corticotropin with a C-terminal proline-glycine-proline extension, a modification intended to slow enzymatic breakdown. The free peptide has a molecular mass near 813.9 daltons. It belongs to the class of ACTH-derived fragments studied for central nervous system activity rather than for adrenal steroid stimulation. This structural relationship to a natural hormone fragment is the usual starting point for describing the compound in the literature.
Development took place during the 1980s at institutes of the Russian Academy of Sciences, where peptide fragments of ACTH were screened for cognitive and neuroprotective effects. The compound received regulatory approval in Russia as a nasal preparation, marketed for conditions such as ischemic stroke, transient ischemic attacks, and optic nerve disorders. Registration in other countries has not followed. Most published clinical reports originate from a small number of Russian research groups, and independent replication outside that setting remains limited. The regulatory status therefore differs sharply between Russia and the rest of the world.
Laboratory work points toward modulation of neurotrophic signaling, particularly expression of brain-derived neurotrophic factor and nerve growth factor in hippocampal tissue. Studies also describe effects on monoamine turnover, inflammatory mediators, and oxidative markers. These observations come mainly from animal models and cultured cells, so the causal chain in humans is not firmly established. Whether the reported molecular changes translate into measurable clinical benefit is an open question. Reviews generally present the mechanism as plausible rather than demonstrated.

=== Occurrence === Glutathione is the most abundant non-protein thiol (R−SH-containing compound) in animal cells, ranging from 0.5 to 10 mmol/L. It is present in the cytosol and organelles. The concentration of glutathione in the cytoplasm is significantly higher (ranging from 0.5-10 mM) compared to extracellular fluids (2-20 μM), reaching levels up to 1000 times greater. In healthy cells and tissue, more than 90% of the total glutathione pool is in the reduced form (GSH), with the remainder in the disulfide form (GSSG). The cytosol holds 80-85% of cellular GSH, and the mitochondria hold 10-15%. Human beings synthesize glutathione, but a few eukaryotes do not, including some members of Fabaceae, Entamoeba, and Giardia. The only known archaea that make glutathione are halobacteria. Some bacteria, such as "Cyanobacteria" and Pseudomonadota, can biosynthesize glutathione. The systemic availability of orally administered glutathione is poor. It has low bioavailability because the tripeptide is the substrate of proteases (peptidases) of the alimentary canal, and due to the absence of a specific carrier of glutathione at the level of the cell membrane. The administration of N-acetylcysteine (NAC), a cysteine prodrug, helps replenish intracellular GSH levels.
== Prevention == Moderate-quality evidence exists that suggests that the combination of education and exercise may reduce an individual's risk of developing an episode of low back pain. Lesser-quality evidence points to exercise alone as a possible deterrent to the risk of the condition.
== Development and current usage == Livia S. Eberlin, Ph.D., an Assistant Professor of Chemistry at the University of Texas at Austin, first reported the invention of the MasSpec Pen in 2017. Within her laboratory, the device has been used to analyze human tissue biopsies, including normal and cancerous breast, lung, ovarian, and thyroid samples. The MasSpec Pen is currently being evaluated for use on freshly excised tissue biopsies and for intraoperative use during oncology surgeries. The MasSpec Pen has been licensed to Genio Technologies, Inc., a subsidiary of MS Pen Technologies, Inc.

In terms of coregulator recruitment, the ratios of coactivators to corepressors vary in different tissues throughout the body, and it is thought that SARMs may have agonistic effects in tissues with an excess of coactivators relative to corepressors like muscle and bone and may have partially agonistic or antagonistic effects in tissues with an excess of corepressors over coactivators like the prostate. Another mechanism may be that SARMs like enobosarm induce the N/C interaction less readily than AR full agonists like DHT. Induction of the N/C interaction has been associated with the effects of endogenous and exogenous AR agonists, for instance virilization and prostate growth. In animal studies, enobosarm has shown potent muscle-promoting effects that were similar to those of testosterone and DHT. In one of the first published studies, enobosarm maximally restored prostate weight to 51%, seminal vesicle weight to 98%, and levator ani muscle weight to 136% in castrated male rats relative to gonadally intact control male rats, with an ED50 dose for muscle of 0.03 mg/day. For comparison, testosterone propionate was able to maximally stimulate levator ani muscle to 104% and prostate weight to 121%, with ED50 doses of 0.15 mg/day and 0.13 mg/day, respectively. Hence, enobosarm was able to stimulate the levator ani muscle to a size greater than that in normal male rats or produced with exogenous testosterone in castrated male rats, but was only capable of partially rescuing prostate gland weight.
== Droplet CCC == Droplet countercurrent chromatography (DCCC) was introduced in 1970 by Tanimura, Pisano, Ito, and Bowman. DCCC uses only gravity to move the mobile phase through the stationary phase which is held in long vertical tubes connected in series. In the descending mode, droplets of the denser mobile phase and sample are allowed to fall through the columns of the lighter stationary phase using only gravity. If a less-dense mobile phase is used it will rise through the stationary phase; this is called ascending mode. The eluent from one column is transferred to another; the more columns that are used, the more theoretical plates can be achieved. DCCC enjoyed some success with natural product separations but was largely eclipsed by the rapid development of high-speed countercurrent chromatography. The main limitation of DCCC is that flow rates are low, and poor mixing is achieved for most binary solvent systems.
Enzymatic degradation of κ-casein. Micellar flocculation. Gel formation. Each step follows a different kinetic pattern, the limiting step in milk-clotting being the degradation rate of κ-casein. The kinetic pattern of the second step of the milk-clotting process is influenced by the cooperative nature of micellar flocculation, whereas the rheological properties of the gel formed depend on the type of action of the proteases, the type of milk, and the patterns of casein proteolysis. The overall process is influenced by several different factors, such as pH or temperature. The conventional way of quantifying a given milk-clotting enzyme employs milk as the substrate and determines the time elapsed before the appearance of milk clots. However, milk clotting may take place without the participation of enzymes because of variations in physicochemical factors, such as low pH or high temperature. Consequently, this may lead to confusing and irreproducible results, particularly when the enzymes have low activity. At the same time, the classical method is not specific enough, in terms of setting the precise onset of milk gelation, such that the determination of the enzymatic units involved becomes difficult and unclear. Furthermore, although it has been reported that κ-casein hydrolysis follows typical Michaelis–Menten kinetics, it is difficult to determine with the classic milk-clotting assay.
=== The California Sterilization Act of 1909 === This law passed in California in 1909, authorized medical staff like doctors and medical superintendents to perform sterilization procedures on both men and women deemed as feeble-minded, whose mental diseases, IQ, and intelligence could be passed down to future generations. A survey in mental deviations in prisons, public schools, and orphanages in California institutions reported worriness of feeble-mindedness and relation of intelligence to previous delinquency record. In their survey, they found that California had drawn a large proportion of immigrants of undesirable types and would therefore recommend them to sterilization processes. Statistical evidence demonstrates that there the number of sterilizations were disproportionate to racial and ethnic minorities, such as people of low class and female gender. Research also suggests that Latina women were targeted at significantly higher rates than non-Latinas. A 2018 study by Novak and colleagues analyzed over 17,000 forms recommending institutionalized patients for sterilization between 1920 and 1945. The researchers found that Latinas had a 59% greater risk of sterilization compared to non-Latinas. These findings of disproportionate sterilization represent how eugenic thinking reinforced racial stereotypes that portrayed Latinas as inferior and unfit to reproduce.

The sequence is Met-Glu-His-Phe-Pro-Gly-Pro, a seven-residue peptide. The first four residues correspond to the ACTH(4-10) fragment, and Pro-Gly-Pro is added at the C-terminus. N-acetylated versions also appear in the literature and differ in mass.
Semax is a synthetic analog of the ACTH(4-10) fragment rather than a copy of the full hormone. Truncation plus the added tail removes the steroidogenic portion of ACTH. That distinction explains why it is studied for nervous-system effects instead of adrenal stimulation.
It is registered as a prescription product in Russia and has been used there clinically, chiefly by the intranasal route. No approval exists in the United States or the European Union. Availability elsewhere typically falls under research-chemical distribution, with varying purity documentation.
