==== Cancer therapeutics ==== In 2022, over 1.9 million new cancer cases are projected in the U.S. alone. Nanogels are an attractive drug delivery solution for increasing both the efficacy of cancer therapeutics and their localization to cancer cells. Nanogels are currently being investigated for the treatment of different types of cancer, of which a few examples are listed here. In one study, chitosan-based nanogels loaded with doxorubicin, a chemotherapeutic, with a positive surface charge demonstrated a lower colorectal cancer cell viability compared to control groups and a similarly loaded nanogel with a negative surface charge. Another group conjugated folic acid to nanogels loaded with cisplatin or doxorubicin and delivered these therapeutics to ovarian cancer cells, which overexpress the folate receptor that binds with folic acid. These conjugated nanogels produced a significant decrease in tumor growth in a mouse model compared to vehicle controls and showed a site-specific delivery model for nanogels that may be effective for other types of cancer with upregulated folate receptors. Interestingly, gelatin-based nanogels loaded with cisplatin and conjugated to epidermal growth factor receptor (EGFR) ligands have been reported to successfully target lung cancer cells both in vitro and in vivo, with additional work confirming the effectiveness of these nanogels when transformed into aerosol particles.

Four additional trials (NCT03131687, NCT03311724, NCT03861052, and NCT03861039) were included in the safety evaluation, for a total of 7,769 adult participants with type 2 diabetes; therefore, the number of participants representing efficacy findings may differ from the number representing safety findings due to different pools of study participants analyzed for efficacy and safety. Tirzepatide's benefits for the treatment of adult participants with type 2 diabetes were primarily evaluated in five clinical trials. In May 2022, the Committee for Medicinal Products for Human Use (CHMP) of the European Medicines Agency (EMA) adopted a positive opinion, recommending the granting of a marketing authorization for the medicinal product Xenpozyme, intended for the treatment of non-central nervous system (CNS) manifestations of acid sphingomyelinase deficiency type A/B or type B. Xenpozyme was reviewed under the accelerated assessment program of the European Medicines Agency (EMA). The applicant for this medicinal product is Genzyme Europe BV. Olipudase alfa was approved for medical use in the European Union in June 2022.
=== Wild birds === Wildlife, including wild and migratory birds, serve as indicators of ecosystem health. They act as a reservoir for zoonotic diseases and antimicrobial-resistant organisms. Birds are a key link between the transmission of zoonotic diseases to human populations. By the same token, increased contact between wild birds and human populations (including domesticated animals), has increased the amount of anti-microbial resistance (AMR) to the bird population. The introduction of AMR to wild birds positively correlates with human pollution and increased human contact. Additionally, wild birds can participate in horizontal gene transfer with bacteria, leading to the transmission of antibiotic-resistant genes (ARG). For simplicity, wild bird populations can be divided into two major categories, wild sedentary birds and wild migrating birds. Wild sedentary bird exposure to AMR is through increased contact with densely populated areas, human waste, domestic animals, and domestic animal/livestock waste. Especially if they are predators, wild, sedentary birds can come into contact with prey that interact more closely with humans. Wild migrating birds interact with sedentary birds in different environments along their migration route. This increases the rate and diversity of AMR across varying ecosystems. Neglect of wildlife in the global discussions surrounding health security and AMR, creates large barriers to true AMR surveillance.
== History == Oxandrolone was first made by Raphael Pappo and Christopher J. Jung while at Searle Laboratories, now part of Pfizer and they first described the drug in 1962. They were immediately interested in oxandrolone's very weak androgenic effects relative to its anabolic effects. It was introduced as a pharmaceutical drug in the United States in 1964. Oxandrolone was prescribed to promote muscle regrowth in disorders which cause involuntary weight loss, and is used as part of treatment for HIV/AIDS. It had also been shown to be partially successful in treating cases of osteoporosis. However, in 1989, in part due to bad publicity from its illicit use by bodybuilders, production of Anavar was discontinued by Searle Laboratories. It was picked up by Bio-Technology General Corporation, which changed its name to Savient Pharmaceuticals. In 1995, following successful clinical trials, Savient released it under the brand name Oxandrin. As of 2011 BTG subsequently had won approvals for orphan drug status by the Food and Drug Administration for treating alcoholic hepatitis, Turner syndrome, and HIV-induced weight loss and as an offset to protein catabolism caused by long-term administration of corticosteroids.

These heteroduplexes will more readily melt at lower temperatures. Hence they are selectively denatured at the Tc. Primer annealing stage. The homo-duplex DNA will preferentially remain double stranded and not be available for primer annealing. Extension stage. The DNA polymerase will extend complementary to the template DNA. Since the heteroduplex DNA is used as template, a larger proportion of minor variant DNA will be amplified and be available for subsequent rounds of PCR. Secretary of State Rubio activated a disaster assistance response team. By October 31, three U.S. CH-47 Chinook helicopters were deployed to Jamaica to help with relief efforts, with five more on the way. === 12-HHT is a BLT2 receptor agonist === Leukotriene B4 (LTB4) is an arachidonic acid metabolite made by the 5-lipoxygenase enzyme pathway. It activates cells through both its high affinity (dissociation constant [Kd] of 0.5–1.5 nM) Leukotriene B44 receptor 1 (BLT1 receptor) and its low affinity BLT2 receptor (Kd=23 nM); both receptors are G protein-coupled receptors that, when ligand-bound, activate cells by releasing the Gq alpha subunit and pertussis toxin-sensitive Gi alpha subunit from heterotrimeric G proteins. BLT1 receptor has a high degree of ligand-binding specificity: among a series of hydroxylated eicosanoid metabolites of arachidonic acid, it binds only LTB4, 20-hydroxy-LTB4, and 12-epi-LTB4; among this same series, BLT2 receptor has far less specificity in that it binds not only LTB4, 20-hydroxy-LTB4, and 12-epi-LTB4, but also 12(R)-HETE and 12(S)-HETE (i.e. the two stereoisomers of 12-hydroxyeicosatetraenoic acid) and 15(S)-HpETE and 15(S)-HETE (i.e. the two stereoisomers of 15-hydroxyicosatetraenoic acid). All of these binding affinities are considered to be low and therefore indicating that some unknown ligand(s) might bind BLT2 with high affinity. === Methemoglobinemia === Methylene blue is used to treat methemoglobinemia by chemically reducing the ferric iron in hemoglobin to ferrous iron. Methemoglobinemia can arise from ingestion of certain pharmaceuticals, toxins, or broad beans in those susceptible. Specifically, it is used to treat methemoglobin levels that are greater than 30% or in which there are symptoms despite oxygen therapy. Normally, through the NADH- or NADPH-dependent methemoglobin reductase enzymes, methemoglobin is reduced back to hemoglobin. When large amounts of methemoglobin occur secondary to toxins, methemoglobin reductases are overwhelmed. Methylene blue, when injected intravenously as an antidote, is itself first reduced to leucomethylene blue, which then reduces the heme group from methemoglobin to hemoglobin. Methylene blue can reduce the half-life of methemoglobin from hours to minutes. At high doses, however, methylene blue actually induces methemoglobinemia, reversing this pathway. Methylene blue is indicated for treating sodium nitrite overdoses, in particular when sodium nitrite is used as part of a suicide attempt.
=== Research applications === Ribosomal proteins: For the studies of individual ribosomal proteins, the use of proteins that are produced and purified from recombinant sources has largely replaced those that are obtained through isolation. However, isolation is still required for the studies of the whole ribosome. Lysosomal proteins: Lysosomal proteins are difficult to produce recombinantly due to the number and type of post-translational modifications that they have (e.g. glycosylation). As a result, recombinant lysosomal proteins are usually produced in mammalian cells. Plant cell culture was used to produce FDA-approved glycosylated lysosomal protein-drug, and additional drug candidates. Recent studies have shown that it may be possible to produce recombinant lysosomal proteins with microorganisms such as Escherichia coli and Saccharomyces cerevisiae. Recombinant lysosomal proteins are used for both research and medical applications, such as enzyme replacement therapy.
